Lorbrexen 100 (Lorlatinib) 100 mg Tablets
Lorbrexen 100 mg (Lorlatinib) is a third-generation ALK and ROS1 tyrosine kinase inhibitor used for the treatment of certain forms of non-small cell lung cancer.
Manufacturer: Everest Pharmaceuticals Ltd.. Each pack contains 30 tablets. It is designed to overcome resistance to earlier ALK inhibitors and has excellent penetration through the blood-brain barrier, making it highly effective against brain metastases.

Lorlatinib is active against many known ALK resistance mutations and is used in patients with ALK-positive advanced NSCLC, including those whose disease has progressed after prior targeted therapy.
Indicated for the treatment of adult patients with Non-Small Cell Lung Cancer (NSCLC):
- ๐น ALK-positive NSCLC: in locally advanced or metastatic disease.
- ๐น After progression on prior therapy: following treatment with other ALK inhibitors.
- ๐น Brain metastases: due to strong central nervous system penetration.
Packaging: 100 mg tablets.
โ ๏ธ ADMINISTRATION PROTOCOL:
- Recommended Dose: 100 mg orally once daily.
- Food Intake: Can be taken with or without food.
- Timing: Take at the same time each day.
- Administration: Swallow whole; do not chew, crush, or split the tablet.
- Monitoring: Lipid profile, ECG, liver function, and neurologic status should be monitored during treatment.
- Hypersensitivity to Lorlatinib or any component of the product.
- Pregnancy and breastfeeding.
- Concomitant use with strong CYP3A inducers.
- Use with caution in patients with cardiac conduction disorders, hyperlipidemia, CNS disorders, or hepatic impairment.
Common and clinically important adverse reactions include:
- ๐ง Neurologic/Psychiatric: Cognitive effects, mood changes, irritability, peripheral neuropathy.
- ๐ Metabolic: Increased cholesterol and triglyceride levels.
- ๐ง General: Edema, weight gain, fatigue.
- โ ๏ธ Cardiac: PR interval prolongation, AV block, decreased left ventricular ejection fraction.
- ๐ฉธ Other: Anemia, elevated liver enzymes, dyspnea.
